CLR325 is an intravenously administered synthetic cyclic peptide and apelin-13 analog developed by Novartis for the treatment of heart failure. It acts as an agonist of the apelin receptor (also known as APJ or APLNR), aiming to enhance cardiac contractility and help the heart pump more blood, similar to the endogenous peptide apelin. The drug was investigated in clinical trials for chronic stable heart failure but has not been approved for any indication. Clinical development reached Phase 2 before being discontinued. Adverse events reported in trials included both non-serious (e.g., headache, nausea) and serious events (e.g., bronchitis, hepatic enzyme increase, cardiac failure congestive). The primary mechanism involves activation of the apelin receptor pathway to improve cardiovascular function[1][2][3][4][7].
