Overview

cLRRC15-IFNa is a conditionally active biologic (CAB) designed to deliver interferon-alpha (IFN-α) signaling specifically to the tumor microenvironment by targeting LRRC15-expressing cancer-associated fibroblasts (CAFs). Developed by Tallac Applied Technologies using their proprietary Dual-Binding Antibody (DBA) platform, the molecule is engineered to remain systemically inactive in circulation. Upon binding to LRRC15 on the surface of myofibroblasts, the IFN-α component becomes active, driving the reprogramming of CAFs to counter TGF-β-mediated immunosuppression and stimulating adjacent immune cells, such as CD8+ T cells and dendritic cells. This approach aims to harness the potent anti-tumor activity of IFN-α while avoiding the severe dose-limiting toxicities associated with systemic cytokine administration.
