CLS-015 is a recombinant human deoxyribonuclease I (rhDNase I) being developed by Tel-Aviv Sourasky Medical Center. It is designed to degrade extracellular DNA, specifically targeting neutrophil extracellular traps (NETs) within the tumor microenvironment. The drug is currently being evaluated in a Phase 1 clinical trial as an adjunctive therapy to anti-CD19 CAR-T cell therapy for patients with stable or progressive large B-cell lymphoma (LBCL) at the time of lymphodepletion. The rationale for this combination is that NETs can create a physical and biochemical barrier that impairs the infiltration and function of CAR-T cells; by degrading these structures, CLS-015 aims to enhance the efficacy and safety of the cellular immunotherapy.
