Drug intelligence / Profile preview
CLS-S1 is a preclinical-stage small molecule dual inhibitor of ADAMTS4 and ADAMTS5 (A Disintegrin and Metalloproteinase with Thrombospondin Motifs 4 and 5) developed by Cellus. By concurrently inhibiting ADAMTS4 and ADAMTS5, CLS-S1 prevents the cleavage of extracellular matrix (ECM) components such as aggrecan and versican, thereby controlling ECM remodeling and regulating the tumor microenvironment (TME). This inhibition blocks the conversion of membrane-bound CXCL16 into its soluble form, which in turn suppresses the recruitment and differentiation of tumor-associated macrophages (TAMs) and enhances CD8+ T cell activation. CLS-S1 is being developed for the treatment of solid tumors, including triple-negative breast cancer.
