CLT030

**CLT030** is an investigational antibody-drug conjugate (ADC) developed for the treatment of acute myeloid leukemia (AML). It consists of a humanized anti-CLL1 monoclonal antibody with two engineered cysteine residues covalently linked via a cleavable linker to a highly potent DNA-binding payload (isoquinolidinobenzodiazepine, IQB, also referred to as D211). The ADC targets C-type lectin domain family 12 member A (CLL1, or CLEC12A), which is highly expressed on leukemic stem cells (LSCs) and AML blasts but absent on normal hematopoietic stem cells (HSCs), enabling selective killing of malignant cells while sparing healthy hematopoiesis. Preclinical data demonstrate potent inhibition of LSC colony formation in vitro, robust tumor regression in AML cell line and patient-derived xenograft models in vivo, and reduced myelotoxicity compared to CD33-targeted ADCs. Developed by Cellerant Therapeutics, CLT030 aims to address AML relapse driven by chemotherapy-resistant LSCs, with plans for IND filing and Phase 1 trials.
