Overview

CLY-101 is an investigational autologous cell therapy developed by Cellerys AG, designed to induce antigen-specific immune tolerance in patients with multiple sclerosis (MS), particularly the relapsing-remitting form (RRMS). The therapy involves the ex vivo chemical coupling of seven distinct myelin-derived peptides—originating from myelin basic protein (MBP), proteolipid protein (PLP), and myelin oligodendrocyte glycoprotein (MOG)—onto the surface of the patient's own red blood cells (erythrocytes) using a cross-linking agent. Upon re-infusion, these modified cells undergo natural senescence and are sequestered by the spleen and liver. The subsequent phagocytosis by antigen-presenting cells in a non-inflammatory environment promotes the expansion of regulatory T cells and the deletion or anergy of pathogenic autoreactive T cells. This mechanism aims to restore immune tolerance and halt the autoimmune attack on the central nervous system without causing generalized immunosuppression.
