CLYM116 is a preclinical-stage, Fc-engineered monoclonal antibody that targets APRIL (A Proliferation-Inducing Ligand, also known as tumor necrosis factor ligand superfamily member 13). It is designed to inhibit APRIL signaling and deplete immunoglobulin A (IgA), addressing pathogenic B-cell activity in autoimmune diseases. The antibody employs a novel pH-dependent bind-and-release mechanism that not only blocks APRIL’s binding to its receptors but also promotes lysosomal degradation of APRIL, resulting in rapid, deep, and durable inhibition of the pathway. This engineering extends serum half-life for improved exposure and less frequent dosing. Developed primarily for IgA nephropathy—a leading cause of glomerulonephritis—CLYM116 may have broader utility across other B-cell-mediated diseases where APRIL plays a critical role[1][2][3][5][6].
