CM-128 is an investigational small-molecule inhibitor of calcium-release activated calcium (CRAC) channels being developed by CalciMedica for the treatment of acute pancreatitis and related inflammatory conditions. By blocking store-operated calcium entry through CRAC channels in non-excitable cells such as pancreatic acinar cells and immune cells, CM-128 is intended to prevent pathological intracellular calcium overload, limit cell injury and necrosis, and attenuate downstream inflammatory cascades that drive disease severity in pancreatitis. The program is at an early clinical development stage, with CalciMedica pursuing proof-of-concept in acute pancreatitis where CRAC channel–mediated Ca2+ dysregulation plays a central pathogenic role.[1]
