Overview

CM-2501 is a humanized monoclonal antibody designed to target the hepatocyte growth factor receptor (c-Met), a receptor tyrosine kinase that is frequently overexpressed, amplified, or mutated in various solid tumors. Developed by CureMab, CM-2501 functions by binding to the extracellular SEMA domain of c-Met, thereby blocking the binding of its natural ligand, hepatocyte growth factor (HGF). This inhibition prevents receptor dimerization and the subsequent activation of downstream oncogenic signaling pathways, including the PI3K/Akt and Ras/MAPK cascades, which drive tumor cell proliferation, survival, and metastasis. In addition to its ligand-blocking activity, CM-2501 induces the internalization and lysosomal degradation of the c-Met receptor, effectively reducing its surface expression. This dual mechanism of action is intended to provide more robust inhibition of c-Met signaling compared to traditional small molecule inhibitors and to overcome certain resistance mechanisms. It is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors.
