CM-ACD3 is a preclinical antibody-drug conjugate (ADC) being developed by Curemeta for the treatment of gastric cancer. The drug consists of a monoclonal antibody targeting the TRA-1-60 epitope, which is a carbohydrate antigen expressed on certain cancer cells and pluripotent stem cells, conjugated to a DNA-alkylating payload. This mechanism of action distinguishes CM-ACD3 from Curemeta's other lead candidates, CM-09 and CM-14, which utilize the microtubule inhibitor monomethyl auristatin E (MMAE). By targeting TRA-1-60, CM-ACD3 delivers its cytotoxic DNA-damaging agent directly to tumor cells, aiming to induce cell death while minimizing systemic toxicity.
