CM5480 is a selective small molecule inhibitor of Orai1, a calcium release-activated calcium (CRAC) channel protein. It is designed to block store-operated calcium entry (SOCE) implicated in cellular pathologies, specifically in pancreatic acinar, ductal, and stellate cells. CM5480 has shown preclinical efficacy in preventing the progression of recurrent acute pancreatitis (RAP) and early chronic pancreatitis (CP) to established CP by reducing intracellular calcium overload, diminishing immune cell infiltration, restoring secretory and enzyme activity, and inhibiting activation and migration of pancreatic stellate cells to reduce fibrosis[1][2][3][5][7][9][10]. The drug has been developed and studied mainly by CalciMedica, often in collaboration with academic and corporate partners.
