Overview
CMAB017 is an **investigational anti-EGFR probody therapeutic** developed for the treatment of advanced solid tumors—including but not limited to colorectal cancer, squamous cell carcinoma of the head and neck, and esophageal squamous cell carcinoma. It utilizes a novel *probody* strategy: a peptide mask blocks the antibody's binding site, designed to be cleaved in the tumor microenvironment, thereby reducing off-tumor toxicity and lowering skin/gastrointestinal adverse reactions compared to conventional anti-EGFR therapies. The antibody is of the human IgG1 isotype, potentially enhancing Fc-mediated anti-tumor effects. CMAB017 is reported to achieve higher local concentration in tumors following administration. It is considered a first-in-class biological drug platform for probody therapeutics, with development led by Mabpharm and Taizhou Mabtech Pharmaceutical[1][2][3][7][9].
