Overview

CMAB820 is a recombinant humanized monoclonal antibody designed to target the epidermal growth factor receptor (EGFR). Developed by Shanghai Zhangjiang Biotech, a subsidiary of Mabpharm, it is being investigated as a biosimilar to necitumumab. The antibody works by binding specifically to the extracellular domain of EGFR, thereby competitively inhibiting the binding of its natural ligands, such as epidermal growth factor (EGF) and transforming growth factor-alpha (TGF-α). This blockade prevents receptor dimerization and the subsequent activation of downstream signaling pathways, including the MAPK and PI3K/Akt cascades, which are critical drivers of tumor cell proliferation, survival, and angiogenesis. CMAB820 is primarily in clinical development for the treatment of advanced non-small cell lung cancer (NSCLC), particularly squamous cell carcinoma, where EGFR signaling is frequently dysregulated.
