Drug intelligence / Profile preview
CMATAC peptide is a chaperone-mediated autophagy-targeting chimera (CMATAC) peptide designed to selectively degrade transmembrane protein 59 (TMEM59). TMEM59 is a type I transmembrane protein that negatively regulates microglial phagocytosis of amyloid-beta (Aβ) by destabilizing triggering receptor expressed on myeloid cells 2 (TREM2). By targeting TMEM59 for lysosomal degradation via chaperone-mediated autophagy, the CMATAC peptide stabilizes TREM2, promoting microglial Aβ phagocytosis, reducing amyloid plaque burden, and mitigating cognitive and behavioral deficits. It is under preclinical investigation for the treatment of Alzheimer's disease and amyloidosis.
