**CMB-401** is an investigational, discontinued **MUC1-targeted antibody-drug conjugate** developed from the humanized anti-polymorphic epithelial mucin antibody hCTM01 linked through an amide linker to the enediyne cytotoxin calicheamicin. The antibody component targets Mucin-1 on epithelial tumor cells, supporting cellular internalization and delivery of calicheamicin, which induces DNA damage. CMB-401 reached phase 2 testing in recurrent platinum-sensitive epithelial ovarian carcinoma, but no partial responses were observed among evaluable patients and further development was discontinued.
