Overview

CMB305 is a first-in-class prime-boost cancer vaccine immunotherapy designed to induce a robust immune response against the NY-ESO-1 tumor antigen. It consists of two components administered sequentially: - LV305: A lentiviral vector engineered to express full-length NY-ESO-1 RNA and target dendritic cells using a mutated Sindbis virus envelope. This primes the immune system by inducing NY-ESO-1–specific CD8 T-cell responses. - G305: A recombinant adjuvanted NY‑ESO‑1 protein combined with glucopyranosyl lipid A in stable emulsion (GLA-SE), which acts as a TLR4 agonist to boost and broaden the immune response by activating antigen-presenting cells. CMB305 was developed primarily for patients with soft tissue sarcomas expressing NY‑ESO‑1 (notably synovial sarcoma and myxoid/round cell liposarcoma). Clinical trials have shown that it can induce both antibody and T-cell responses against NY‑ESO‑1. The regimen has been studied alone and in combination with checkpoint inhibitors such as atezolizumab[2][3][5][6].
