CMD-193 is an antibody-drug conjugate (ADC) composed of a humanized monoclonal antibody (Hu3S193) targeting the Lewis Y (LeY) antigen, conjugated to the cytotoxic agent calicheamicin. The LeY antigen is expressed on various tumor cells. Calicheamicin is a potent enediyne antibiotic that induces DNA double-strand breaks, leading to cell death. CMD-193 was developed for its potential antineoplastic activity in cancers expressing the LeY antigen. The drug was initially developed by Wyeth and reached phase 1 clinical trials for advanced solid tumors but development has since been discontinued[1][2][3][6].
