Overview

cMET CAR T cells are an autologous, RNA-electroporated chimeric antigen receptor (CAR) T-cell therapy designed to target the cell-surface antigen cMET. The CAR construct features an extracellular single-chain antibody variable fragment (scFv) derived from the 5D5 antibody, an IgG4 hinge, a human CD8α transmembrane domain, and intracellular signaling domains composed of CD3ζ and 4-1BB costimulatory modules. This therapy was investigated in a pilot phase I trial for metastatic melanoma and metastatic triple-negative breast cancer (mTNBC), administered intravenously without lymphodepleting chemotherapy. The RNA electroporation ensures transient CAR expression, intended as a safety feature.
