Overview

CMK-389 is a fully human, high-affinity IgG1-LALA monoclonal antibody developed by Novartis for the treatment of chronic pulmonary sarcoidosis and atopic dermatitis. Its mechanism of action is the selective inhibition of interleukin 18 (IL-18), a pro-inflammatory cytokine implicated in TH1-polarized immune responses and granuloma formation in sarcoidosis. By neutralizing IL-18, CMK-389 aims to reduce tissue damage associated with excessive IFN-gamma-driven immune responses. The drug has been evaluated in phase II clinical trials for both pulmonary sarcoidosis and atopic dermatitis but did not demonstrate meaningful clinical benefit in pulmonary sarcoidosis compared to placebo[1][2][5][6].
