Overview

CMPD 167 is a small molecule antagonist of the C-C chemokine receptor type 5 (CCR5), originally developed by Merck & Co., Inc. for the treatment and prevention of HIV-1 and Simian Immunodeficiency Virus (SIV) infections. By binding to the CCR5 coreceptor on the surface of host cells, the drug prevents the viral envelope protein from interacting with the receptor, thereby blocking viral entry. Preclinical studies in macaques demonstrated its efficacy in reducing plasma viremia and its potential as a topical microbicide when formulated in vaginal gels or rings. Although it showed significant antiviral activity in animal models, development for human clinical use was discontinued, and the compound currently serves primarily as a pharmacological research tool in the study of viral entry and chemokine receptor signaling.
