CMS-D003

CMS-D003 is a small molecule cardiac myosin inhibitor being developed by Shenzhen Kangzhe Pharmaceutical, a subsidiary of China Medical System Holdings. The drug targets and inhibits cardiac myosin adenosine triphosphate (ATP) enzymes, which reduces myosin-actin interactions and myofilament sliding. This mechanism inhibits myocardial hypercontractility, a hallmark of hypertrophic cardiomyopathy (HCM), particularly obstructive hypertrophic cardiomyopathy (oHCM). By reducing over-contraction, CMS-D003 aims to improve diastolic dysfunction. The drug is designed with a short half-life and a favorable safety profile, including minimal risk for drug-drug interactions. It received Investigational New Drug (IND) approval in March 2025 and is currently in Phase I clinical trials to evaluate safety, tolerability, and pharmacokinetics. Future development may include treatment for heart failure with preserved ejection fraction (HFpEF).
