The CMTM6 nanobody is a therapeutic agent designed to target the chemokine-like transmembrane protein 6 (CMTM6). CMTM6 plays a crucial role in stabilizing various membrane proteins, including the Epidermal Growth Factor Receptor (EGFR) and Programmed Death-Ligand 1 (PD-L1), by preventing their lysosomal degradation. In non-small cell lung cancer (NSCLC), the nanobody works by blocking the interaction between CMTM6 and EGFR, which leads to a reduction in EGFR levels and inhibits the proliferation of EGFR-tyrosine kinase inhibitor (TKI)-resistant NSCLC cells. Additionally, the CMTM6 nanobody interferes with CMTM6's ability to stabilize PD-L1, thereby enhancing CD8+ T-cell-mediated cytotoxicity and promoting tumor apoptosis, suggesting its potential in cancer immunotherapy. Preclinical studies have shown its efficacy in overcoming TKI resistance in NSCLC and in inhibiting tumor growth in models of colon cancer and glioblastoma. Developers involved in its study include Peking University International Cancer Institute and the Shanghai Institute of Materia Medica, Chinese Academy of Sciences.
