CMV pp65 peptide refers to synthetic peptides derived from the 65 kDa phosphoprotein (pp65), also known as UL83, of human cytomegalovirus (HCMV). These peptides are commonly used in research and clinical settings to stimulate and study immune responses—particularly T-cell responses—against HCMV. The most widely used format is a pool of overlapping 15-mer peptides spanning the entire sequence of the pp65 protein. This pool is designed to activate both CD4+ and CD8+ T-cells across diverse MHC backgrounds by presenting multiple epitopes recognized by T-cell receptors[2][3][7][9]. Mechanistically, these peptides act as antigens that are processed and presented on MHC molecules by antigen-presenting cells, leading to activation of HCMV-specific T-cells which can produce cytokines such as IFN-γ, IL-2, and TNF[2]. The CMV pp65 protein itself plays a role in viral immune evasion by interfering with interferon signaling pathways[7]. In clinical research contexts, these peptides are used for immunomonitoring or as components in experimental vaccines or cell therapies targeting HCMV infection or certain cancers[1][8].
