The CMV pp65 peptide-loaded Alpha-type-1 polarized dendritic cell vaccine (CMV-alphaDC1) is a therapeutic cancer and infectious disease vaccine. It consists of autologous dendritic cells that have been ex vivo polarized to an alpha-type-1 phenotype and loaded with peptides derived from the cytomegalovirus (CMV) antigen pp65. The alpha-type-1 polarization enhances the ability of these dendritic cells to stimulate robust cytotoxic T lymphocyte responses. Upon administration, the vaccine aims to induce or boost a targeted immune response against cells expressing the CMV pp65 antigen—either in the context of viral infection (such as post-allogeneic hematopoietic stem cell transplantation) or in tumors where this antigen is present (e.g., glioblastoma). The approach leverages both cellular immunotherapy and vaccination principles by using patient-derived immune cells as vehicles for antigen presentation[7][2][10].
