A combination cell therapy consisting of two components: - Dendritic cells (DCs) that have been pulsed with the pp65 antigen from human cytomegalovirus (CMV), and - Allogeneic natural killer (NK) cells. This approach is designed as an immunotherapeutic strategy for cancer, particularly glioblastoma. The CMV-pp65-pulsed dendritic cells act as a vaccine to stimulate an immune response against tumor-associated antigens, while the allogeneic NK cells provide direct cytotoxic activity against tumor or virally infected targets. The use of KIR-HLA-mismatched purified allogenic NK cells enhances anti-tumor efficacy by overcoming inhibitory signals in the tumor microenvironment. This combination has shown durable complete responses in case studies of recurrent glioblastoma without significant toxicity[1][2][5]. Mechanistically, DCs present viral peptides to both T-cells and memory-like adaptive NK (aNK) cell populations, enhancing their proliferation and functional activity against target tumors[5].
