Overview

CMV pp65-specific T-cell therapy is an adoptive cellular immunotherapy developed by Leiden University Medical Center (LUMC) for the treatment of cytomegalovirus (CMV) complications following allogeneic hematopoietic stem cell transplantation (HSCT). The therapy consists of donor-derived (or patient-derived) CD8+ T-cell lines generated ex vivo by stimulating peripheral blood mononuclear cells with HLA-restricted overlapping synthetic peptides spanning the CMV pp65 protein (UL83). These cells are enriched for interferon-gamma production using the CliniMACS Cytokine Capture System and expanded with autologous feeders and interleukin-2. By restoring CMV-specific immunity, the therapy aims to clear CMV viremia and prevent or treat CMV disease in immunocompromised patients. Clinical evaluations at LUMC have demonstrated the therapy's safety and enduring efficacy in clearing CMV infection without inducing graft-versus-host disease.
