Overview

Cytomegalovirus pp65-specific T-cells are a form of adoptive cell therapy consisting of cytotoxic T lymphocytes (CTLs) that specifically recognize the phosphoprotein pp65 antigen from human cytomegalovirus (CMV). These cells are generated by stimulating donor or patient-derived peripheral blood mononuclear cells with overlapping peptide libraries spanning the entire sequence of CMV pp65, leading to expansion and enrichment of both CD4+ and CD8+ T-cell populations reactive to this viral antigen. The primary mechanism is direct recognition and killing of CMV-infected cells via their specificity for the viral pp65 protein. This therapy is primarily developed for patients with refractory or persistent CMV viremia, especially after allogeneic hematopoietic stem cell transplantation, where conventional antiviral drugs may fail or cause toxicity. The approach can use third-party donor-derived cells when matched donors are unavailable[1][2][4][8].
