Overview

CMX182 is an experimental oral antiviral prodrug developed by Chimerix, Inc., utilizing their proprietary ProLipTag lipid prodrug technology. The compound is a lipid-nucleoside conjugate, specifically a hexadecyloxypropyl (HDP) derivative of a nucleoside analog (often identified as lamivudine/3TC), designed to enhance oral bioavailability and facilitate targeted delivery to the liver. By mimicking natural lipids, CMX182 is absorbed via the intestinal lymphatic system, which bypasses first-pass metabolism and reduces systemic toxicity. Once inside target cells, the lipid moiety is cleaved to release the active nucleoside monophosphate, which is subsequently phosphorylated to its active triphosphate form. This active metabolite inhibits viral DNA polymerase, thereby terminating viral DNA chain elongation. CMX182 was primarily investigated for the treatment of chronic Hepatitis B Virus (HBV) infection but was discontinued in early development as Chimerix prioritized other candidates in its antiviral pipeline.
