CN1 + CN401 is a combination therapy developed by Curon Biopharmaceutical for the treatment of relapsed or refractory lymphoid malignancies. CN1 is a recombinant humanized bispecific monoclonal antibody that simultaneously targets **CD47** and **Programmed cell death 1 ligand 1 (PD-L1)**. It is designed to block the CD47-SIRPα "don't eat me" signal and the PD-1/PD-L1 immune checkpoint, thereby enhancing macrophage-mediated phagocytosis and T-cell activation against tumor cells. CN401 is a small molecule dual inhibitor of **phosphatidylinositol 3-kinase gamma (PI3Kγ)** and **delta (PI3Kδ)** isoforms, which are critical for the survival of malignant lymphocytes and the function of immunosuppressive cells in the tumor microenvironment. The combination was evaluated in a Phase I/II clinical trial (NCT04880564) for patients with peripheral T-cell lymphoma (PTCL), cutaneous T-cell lymphoma (CTCL), and B-cell non-Hodgkin's lymphoma (NHL), although the study was terminated by the sponsor.
