Overview

CN54gp140 + GLA-AF is an investigational HIV vaccine candidate consisting of a recombinant trimeric envelope glycoprotein (CN54gp140) and a synthetic adjuvant (GLA-AF). The CN54gp140 component is derived from the HIV-1 clade C 97CN54 strain and is designed to elicit systemic and mucosal immune responses, including binding antibodies and T-cell activity, against the HIV envelope. The adjuvant, glucopyranosyl lipid A-aqueous formulation (GLA-AF), is a Toll-like receptor 4 (TLR4) agonist that enhances the vaccine's immunogenicity by stimulating innate immune pathways. Developed by Imperial College London and its collaborators, this vaccine has been evaluated in Phase I clinical trials, often as part of heterologous prime-boost strategies involving DNA or viral vectors to prevent HIV-1 infection in healthy volunteers.
