CNC-ala-17-E2 is an experimental hormone-linked nitrosourea designed for the targeted treatment of breast cancer. It consists of a cytotoxic nitrosourea moiety, 1-(2-chloroethyl)-1-nitrosocarbamoyl-L-alanine (CNC-ala), covalently linked to estradiol at the 17-position. This design aims to utilize the estrogen receptor (ER) to selectively deliver the DNA-alkylating nitrosourea to ER-positive tumor cells, thereby increasing local concentration and reducing systemic toxicity. Research has demonstrated its activity in various preclinical models, including MXT mammary carcinoma in mice and MNU-induced mammary carcinoma in rats, although its efficacy in human primary breast carcinoma samples in vitro has shown variability.
