CNKSR1 siRNA is a small interfering RNA designed to silence the expression of the Connector Enhancer of Kinase Suppressor of RAS 1 (CNKSR1) protein. CNKSR1 is a scaffold protein located in the RAS membrane signaling nanocluster, where it facilitates mutant KRAS signaling. By knocking down CNKSR1, the siRNA selectively inhibits the growth and signaling of mutant KRAS-driven cancer cells, such as non-small cell lung cancer (NSCLC) and pancreatic cancer, while sparing cells with wild-type KRAS. This approach targets the dependency of oncogenic KRAS on the CNKSR1 scaffold for its downstream proliferative effects. In research settings, it has been used to validate CNKSR1 as a druggable target for small molecule development.
