Drug intelligence / Profile preview
CNNM4 siRNA is a hepatocyte-targeted small interfering RNA (siRNA) therapeutic designed to silence the expression of Cyclin M4 (CNNM4), a magnesium exporter that is upregulated in various liver pathologies. Developed by Gibela Therapeutics (a spin-off from CIC bioGUNE) in collaboration with Silence Therapeutics using their proprietary mRNAi GOLD platform technology, the siRNA is conjugated to N-acetylgalactosamine (GalNAc) to enable specific delivery to hepatocytes via the asialoglycoprotein receptor (ASGPR). By silencing CNNM4, the therapy restores intracellular magnesium homeostasis, which in turn alleviates mitochondrial dysfunction, reduces endoplasmic reticulum (ER) stress, and mitigates liver injury. CNNM4 siRNA is currently in preclinical development for multiple liver-related conditions, including alpha-1 antitrypsin deficiency (AATD), metabolic dysfunction-associated steatotic liver disease (MASLD/MASH), cholangiocarcinoma (CCA), alcohol-related liver disease (ALD), drug-induced liver injury (DILI), and cholestatic liver disease.
