Overview

CNP-103 is a first-in-class, immune-modifying biodegradable nanoparticle therapy being developed for the treatment of type 1 diabetes. It encapsulates four recombinant proteins—preproinsulin, GAD65, IGRP, and ZnT8—which are key autoantigens implicated in the autoimmune destruction of pancreatic islet cells in type 1 diabetes. By delivering these antigens within nanoparticles to antigen-presenting cells (APCs), primarily in the spleen and liver, CNP-103 aims to induce immune tolerance through reprogramming APCs to present these antigens with a tolerogenic signal. This process leads to T cell anergy or deletion and promotes regulatory T cell (Treg) responses while downregulating pathogenic CD4+ and CD8+ T cell activity. The goal is to prevent further islet cell destruction, maintain endogenous insulin production, and potentially reverse dysglycemia in newly diagnosed or recent-onset type 1 diabetes patients[1][2][4][7].
