CNP-106 is an investigational antigen-specific immune tolerance therapy developed for the treatment of generalized myasthenia gravis. It consists of biodegradable nanoparticles that encapsulate acetylcholine receptor (AChR) epitopes. Upon intravenous administration, these nanoparticles are taken up by antigen-presenting cells such as splenic marginal zone macrophages and liver APCs, leading to tolerogenic immune reprogramming. The mechanism involves inducing anergy or conversion of autoreactive T cells into antigen-specific regulatory T cells (Tregs), thereby downregulating AChR-specific T cell populations and antibodies. This targeted approach aims to address the underlying autoimmune pathology in myasthenia gravis without broad immunosuppression, potentially offering disease-modifying effects with a favorable safety profile[1][3][5][6].
