Overview

CNP-53 is a 53-amino acid isoform of the C-type natriuretic peptide (CNP), which is a member of the natriuretic peptide family and a critical regulator of endochondral bone growth. It is the predominant form of CNP in the central nervous system and certain peripheral tissues. CNP-53 acts as a potent agonist for the Natriuretic Peptide Receptor 2 (NPR2), also known as Guanylate Cyclase B (GC-B). Binding to NPR2 increases intracellular cyclic guanosine monophosphate (cGMP) levels, which in turn inhibits the MAPK/ERK signaling pathway downstream of Fibroblast Growth Factor Receptor 3 (FGFR3). This mechanism is of significant therapeutic interest for skeletal dysplasias such as achondroplasia, where overactive FGFR3 signaling inhibits longitudinal bone growth. Research in CNP-deficient animal models has demonstrated that exogenous administration of CNP-53 can restore craniofacial morphology, increase foramen magnum size, and improve skeletal growth, highlighting its potential as a therapeutic agent for conditions characterized by impaired bone development.
