Overview

CNP-ELP is a long-acting recombinant fusion protein developed by PhaseBio Pharmaceuticals for the treatment of achondroplasia. It consists of a C-type natriuretic peptide (CNP) analogue fused to PhaseBio's proprietary Elastin-Like Polypeptide (ELP) biopolymer. This fusion technology is designed to extend the circulating half-life of the peptide, potentially allowing for weekly or less frequent subcutaneous administration. CNP-ELP acts as an agonist for the Natriuretic peptide receptor B (NPR-B), which increases intracellular cyclic guanosine monophosphate (cGMP) levels. This signaling pathway antagonizes the overactive fibroblast growth factor receptor 3 (FGFR3) pathway, which is the primary driver of growth inhibition in achondroplasia. By restoring the balance between these pathways, CNP-ELP aims to promote endochondral bone growth and improve skeletal development in affected individuals.
