Drug intelligence / Profile preview
CNP-MOG35-55 is a tolerogenic immune-modifying nanoparticle (TIMP) developed by Cour Pharmaceutical Development Company. It consists of biodegradable poly(d,l-lactic-co-glycolic acid) (PLGA) nanoparticles, approximately 400-800 nm in diameter, which encapsulate the myelin oligodendrocyte glycoprotein (MOG) peptide fragment residues 35-55. The drug is designed to treat autoimmune diseases, specifically multiple sclerosis, by inducing antigen-specific immune tolerance. Upon intravenous administration, the negatively charged nanoparticles are selectively captured by MARCO-expressing myeloid cells in the splenic marginal zone and liver. This uptake triggers myeloid cell apoptosis and activates the cGAS/STING pathway through the generation of oxidized DNA (8-OHG). This signaling cascade leads to the local secretion of type I interferons and the subsequent expansion of antigen-specific FoxP3+ regulatory T (Treg) cells and Tr1 cells, which suppress autoreactive T cell responses without systemic immunosuppression.
