Overview

CNP-OVA323-339 is a tolerogenic immune-modifying particle (TIMP) developed by Cour Pharmaceutical Development Company. It consists of biodegradable poly(d,l-lactic-co-glycolic acid) (PLGA) nanoparticles, approximately 400-800 nm in diameter, encapsulating the ovalbumin-derived peptide OVA323-339. The drug is designed to induce antigen-specific immune tolerance by mimicking the natural process of apoptotic cell clearance. Upon intravenous administration, the negatively charged nanoparticles are selectively taken up by MARCO-expressing macrophages and dendritic cells in the spleen and liver. This uptake triggers myeloid cell apoptosis and activates the cGAS/STING pathway, leading to the production of type I interferons (IFN-β). This signaling environment promotes the expansion of antigen-specific FoxP3+ regulatory T cells (Tregs) and Tr1 cells, while inducing anergy in effector T cells. CNP-OVA323-339 is primarily utilized in preclinical research to demonstrate the efficacy of the CNP platform in models of delayed-type hypersensitivity and other immune-mediated conditions.
