Overview

CNS-11g is a small molecule analog of CNS-11 developed as an inhibitor of alpha-synuclein aggregation. It is capable of disaggregating preformed alpha-synuclein fibrils in vitro, preventing the intracellular seeded aggregation of alpha-synuclein, and reducing alpha-synuclein-related cytotoxicity in neuronal cells. Both CNS-11 and CNS-11g have been shown to disassemble fibrils extracted from multiple system atrophy (MSA) patient brains, prevent fibril seeding, and reduce alpha-synuclein aggregates in vivo in animal models. CNS-11g has demonstrated brain penetration in mice and exhibits improved properties over the parent compound with regard to preventing the seeding of alpha-synuclein fibrils, suggesting potential as a therapeutic candidate for synucleinopathies such as Parkinson's disease and multiple system atrophy[1][3][5][7][9].
