Overview

CNTO 0007 is a humanized monoclonal antibody developed by Centocor (now part of Janssen Pharmaceutical) that acts as a potent agonist of the erythropoietin receptor (EPOR). Unlike recombinant human erythropoietin (rhEPO), which is a glycoprotein hormone, CNTO 0007 is an antibody-based mimetic designed to bind and activate the EPOR signaling pathway. The drug was primarily investigated for its potential neuroprotective properties in the treatment of acute ischemic stroke (cerebral infarction). The therapeutic rationale was based on the observation that EPOR activation in the central nervous system can reduce neuronal apoptosis, diminish inflammatory responses, and promote tissue repair following hypoxic or ischemic injury. Clinical development reached Phase 1 testing to evaluate safety and tolerability in stroke patients, but the program was subsequently discontinued.
