**CNTO-530** is a novel erythropoiesis-stimulating agent developed by Centocor using the MIMETIBODY™ platform. It is a 58 kDa dimeric fusion protein consisting of erythropoietin mimetic peptide (EMP1) sequences fused to an antibody Fc domain, with no sequence homology to native erythropoietin (EPO). CNTO-530 acts as a potent, long-acting agonist of the **erythropoietin receptor (EPOR)**, uniquely promoting sustained erythropoiesis by enhancing formation of PODXL-high CD71-high erythroblasts and expanding bone marrow-resident Kit-negative CD71-high Ter119-negative progenitors. It demonstrates superior duration of action compared to epoetin alfa and darbepoetin alfa, maintaining elevated hematocrit, reticulocytes, and hemoglobin for up to 25-28 days after a single dose in preclinical models of steady-state erythropoiesis, chemotherapy-induced anemia (e.g., 5-FU, carboplatin-paclitaxel), anemia of chronic disease, and β-thalassemia. Additional effects include improved glucose tolerance and insulin sensitivity in obese diabetic models via increased glucose uptake in skeletal muscle and heart.[1][2][3][4][5][7][8]
