CNTO3157 is a fully human monoclonal antibody that acts as an antagonist of Toll-like receptor 3 (TLR3). It was developed to target inflammatory pathways triggered by viral infections such as human rhinovirus (HRV), which are known to precipitate asthma exacerbations. By binding the extracellular domain of TLR3 and blocking its activation by double-stranded RNA, CNTO3157 aims to reduce immune-mediated inflammation associated with respiratory viral infections. The drug has been investigated primarily for the prevention or attenuation of asthma exacerbations induced by HRV infection. Clinical studies have evaluated its safety and efficacy in both healthy volunteers and patients with mild-to-moderate asthma[1][2][5]. The drug was originally developed by Centocor Ortho Biotech (now Janssen Biotech) in collaboration with MorphoSys, with further development involving Provention Bio[1].
