Overview

CNTX-3001 is a novel, non-opioid, highly potent and highly selective small molecule agonist of the nociceptin receptor (NOPR or OOR), currently in clinical development as an intrathecal therapy for severe chronic pain, including intractable low back pain. Unlike opioids, CNTX-3001 acts specifically on spinal NOPR to induce powerful analgesia while avoiding risk of addiction and behavioral side effects. It is intended primarily for delivery directly to the spinal cord via lumbar puncture or implantable pump, leveraging its favorable solubility properties for local CNS administration. Preclinical studies have shown superior efficacy and side effect profiles compared to intrathecal morphine, and first-in-human Phase 1 trials are underway[1][2][5][7][8].
