Overview

CNV-2197944 is a novel, orally available small molecule that acts as a state-dependent blocker of the N-type voltage-gated calcium channel Cav2.2 (alpha 1B subunit). It was initially developed for the treatment of chronic pain conditions, with primary clinical investigation in neuropathic pain such as diabetic peripheral neuropathy. The drug selectively inhibits highly active Cav2.2 channels, which are involved in pain signaling by mediating calcium entry into neurons and facilitating neurotransmitter release. By targeting these channels in their activated or inactivated states, CNV-2197944 aims to reduce neuronal hyperexcitability associated with chronic pain while minimizing side effects seen with less selective agents. Clinical trials have reached Phase 2 for diabetic peripheral neuropathy and post-herpetic neuralgia[1][3][6][7].
