CO-1 is a novel bifunctional anti-CD47 monoclonal antibody developed by Caedo Oncology. It is designed to block the interaction between CD47 on cancer cells and SIRPα on myeloid cells, thereby neutralizing the "don't eat me" signal and promoting macrophage-mediated phagocytosis. Distinctively, CO-1 also induces direct, rapid programmed cell death (PCD) in cancer cells through CD47 ligation, a mechanism that is independent of the immune system's effector cells. The antibody is an IgG4 isotype, which helps minimize binding to red blood cells and reduces the risk of hemagglutination. Caedo Oncology is developing CO-1 and its derivatives, CO-001 (a chimeric version) and CO-005 (a humanized single-chain fragment variable-fragment crystallizable fusion protein), for the treatment of hematologic malignancies and potentially other oncology indications.
