co-hu-HSAN-PBD is a novel antibody-drug conjugate (ADC) designed to target GDNF family receptor alpha 2 (GFRA2), a lineage-restricted oncoprotein highly expressed in pediatric solid tumors such as neuroblastoma and Ewing sarcoma. The ADC consists of a humanized version of the murine HSAN antibody (co-hu-HSAN) conjugated to a pyrrolobenzodiazepine (PBD) dimer cytotoxic payload via a cleavable linker. Upon binding to GFRA2 on the tumor cell surface, the ADC is internalized, and the PBD payload is released, leading to DNA cross-linking, double-strand breaks, and subsequent cell death. Preclinical studies have demonstrated potent and specific cytotoxicity in GFRA2-positive cell lines and patient-derived xenograft models, with minimal activity in GFRA2-null tissues.
