Drug intelligence / Profile preview
ALPHATRIM FORTE is an oral fixed-dose combination of the sulfonamide antibiotic sulfamethoxazole and the dihydrofolate reductase inhibitor trimethoprim, collectively known as co-trimoxazole. Developed by Aleppo Pharmaceutical, it is approved for the treatment of various bacterial infections, including urinary tract infections, bronchitis, and middle ear infections. Sulfamethoxazole acts as a competitive inhibitor of dihydropteroate synthase (DHPS), blocking the conversion of para-aminobenzoic acid (PABA) to dihydropteroate. Trimethoprim inhibits bacterial dihydrofolate reductase (DHFR), preventing the reduction of dihydrofolate to tetrahydrofolate. Together, these two agents exert a synergistic bactericidal effect by sequentially blocking two consecutive steps in the bacterial biosynthesis of essential nucleic acids and proteins.
