coAAV-SCS-01 is a novel gene therapy vector developed by Coave Therapeutics using its proprietary ALIGATER™ (Advanced Vectors-Ligand Conjugates) platform. It represents the world's first conjugated AAV capsid specifically engineered for suprachoroidal delivery to treat retinal vascular diseases. The vector is designed through chemical conjugation of targeting ligands to AAV capsids, enabling enhanced tissue specificity and improved safety profiles. In non-human primate studies, coAAV-SCS-01 demonstrated up to 26-fold superior performance compared to first and second-generation AAV capsids (including AAV2, AAV3B, AAV.7m8, AAV.v128, and AAV8) in targeting RPE-choroid and retinal cells. The vector shows de-targeting from off-target areas such as the iris-ciliary body and anterior segment of the eye, along with immune evasion properties that suggest potential for safe second-eye treatment. coAAV-SCS-01 is optimally suited for treating retinal vascular diseases such as neovascular age-related macular degeneration (nAMD) and diabetic macular edema (DME) through suprachoroidal administration, which provides a quick, in-office procedure comparable to routine anti-VEGF injections rather than requiring invasive surgical procedures.
