Overview

**Cobimetinib + niraparib** is a combination of two small molecule inhibitors investigated for the treatment of advanced platinum-sensitive ovarian cancer. *Cobimetinib* is a selective MEK (mitogen-activated protein kinase kinase) inhibitor that blocks the MAPK/ERK signaling pathway, which is often dysregulated in cancer. *Niraparib* is a poly(ADP-ribose) polymerase (PARP) inhibitor that impedes DNA repair in cancer cells, especially those defective in homologous recombination repair, such as cells with BRCA mutations. The combination aims to enhance antitumor activity by simultaneously inhibiting tumor cell proliferation and survival mechanisms. This combination has been studied in a Phase 1 clinical trial (NCT03695380) for patients with advanced platinum-sensitive ovarian cancer, both with and without an additional agent (atezolizumab)[1][2][3][4][6].
